The history of Chinese Immigration to USA - Research Paper Example Wherever they went, the Chinese immigrants brought with them their language, culture, social institutions, and customs; and as time went on, they made lasting contributions to the United States as they tried to become an integral part of the U.S. population. (â€Å"Chinese immigration to the US†) Privation of the Chinese immigrants to USA in the 18th century Integration has not been an easy one because the Chinese underwent a long period of discrimination from their adopted country. At the outset, they were at once discriminately treated from among the European immigrants and other racial minorities. They were the first immigrant group singled out for denial of citizenship by the U.S. Congress in 1882. Chinese immigration is separated into three periods: 1849 – 1882, 1882-1965 and 1965 to present. The first period of immigration that begun with the California Gold rush in 1848 (Golden Venture) was cut short by the Chinese Exclusion Act of 1882. The Exclusion Act suspend ed labor immigration from China for ten years (USA Chronology). During this first period, the Chinese enjoyed the freedom to travel and become contract laborers in America. Thousands of young male peasants from China were recruited to work in the mines, toil the fields as agricultural workers, and become construction workers to construct a vast railroad network, reclaim swamp-lands, build irrigation system, develop the fishing industry, and work in the labor intensive manufacturing industries in the western states. All along, these Chinese workers felt the anti-sentiments of their presence since they were precluded on becoming permanent settlers. Because of this sentiment, the Chinese immigrants kept to themselves an intention of going back to China when the time comes to enjoy the benefit of their retirement, and to advance their own well being while they are in U.S. According to estimates, there were about 110,000 Chinese populations during the first wave of immigration. The secon d wave of immigration showed a very limited number of Chinese immigrants to U.S. This is caused by political tensions between the U.S and the Chinese governments that further curtailed the chance of Chinese immigrants to enter America. Because America felt Chinese laborers were no longer needed, beginning in 1882, the naturalization and immigrations became very strict to them. At the onset of 1882, an economic hardship was also felt in the U.S., so much so that European Americans also had to compete with the works reserved for the Chinese. This era is also characterized by the beginning of racial discrimination and hatred because of competition and anti-Chinese riots were frequent scenes in the streets of San Francisco. These scenarios put pressure on the U.S. government so that the exclusion act was promulgated (The Library of Congress). A quota allowing only 108 Chinese immigrants was set; so from the period 1888 to 1965, only diplomats, merchants and students and their dependents were allowed to enter the U.S (â€Å"Chinese Americans†) Even those Chinese, who are already in the U.S., are largely discriminated upon, and had to confine their presence in segregated places called Chinatown. A Classic Shakespearean Comedy Essay The Chicago Shakespeare Theatre at Navy Pier manages to combine an extravagant and lavish, while at the same time still very intimate, theatre setting with professional acting performances in every production shown. However, the production of As You Like on March 5, 2011 perhaps best exemplifies these significant qualities crucial to Shakespeare plays. Directed by, Gary Griffin, the production is able to captivate the entire audience for the complete duration of the play through its brilliant illustration of spectacle as well as strong narration. However, the most significant factor of the production would undeniably be its inclusion of an alliance of narrative, dramatic, and spectacle elements all intertwined in the overall production. Griffin depicts a traditional Shakespearean comedy with the use of vivid set design and perfect actor selection in the production of As You Like It in a way that separates the comedic production from any other previous Shakespeare play I have seen. The production does so by being able to masterfully combine Shakespearean comedic elements with deeper connotations about self-identity and the power of love crucial to all productions of As You Like It. The thematic elements prevalent throughout the production are contemporary in regards to all Shakespearean comedies in that the play revolves around mistaken identity, the many forms of love, and the resolutions of all misunderstandings in the end of the play. However, the way in which Griffin’s overall production is able to enthrall the audience through brilliant acting, audience inclusion, and beautiful theatre design in such a way that the audience isn’t forced to exert much personal imagination makes As You Like It a classic example of an ideal Shakespearean production. The Chicago Shakespeare Theatre, the producing theatre for director Gary Griffin’s adaptation of As You Like It, is a professional theatre venue and company located on 800 East Grand Avenue in Navy Pier. The theatre is ornately designed with the intent to transform the audience’s connotation of setting and to make them believe/ give the appearance that they are in an actual Shakespearean Theatre without the use of their imagination. The theatre space used for the production of As You Like It is called Jentes Family Auditorium and features a total of 510 seats on three levels surrounding the courtyard theatre’s â€Å"thrust stage†. This specific theatre was modeled/ designed with inspiration coming directly from the Royal Shakespeare Company’s Swan Theatre, evident at first glance, and thus creates an immediate intimate relationship between the actors, stage/ set design, and the entire audience seated around the three sides of the stage. While part of the set is elevated as a â€Å"stage†, the entire area/ space of the theatre is used by the actors throughout the production; when this is incorporated with the fact that no seat is more than thirty feet from the stage, a feeling of intimacy between actors and audience is ever-present. For instance, the audience is engaged into the action of the play immediately by the way in which the stage extends through the middle of the first-floor seating allowing the appearance/ feeling of the actors walking directly out into the middle of the audience members. In an analysis of the overall production of As You Like It it is evident that even though the set, stage, and use of props and vivid images are riveting, the inclusion of an â€Å"all-star ensemble† of some of Chicago’s finest actors is truly the underlying nature in exemplifying the statement that the production is indeed Shakespeare’s â€Å"glorious romantic comedy†. The actors play courtiers, clowns, philosophers, and locals all who are under the classic comedic spell of the trials and conquests of love as the main character, Rosalind, must disguise herself as a boy in order to escape the royal court. The characters, following Rosalind’s lead, venture into the timeless Forest of Arden where Rosalind finds herself trapped by love and mistaken identity in her attempt to teach the man she loves how to win the women of his dreams. A traditional element present in the production of Shakespearean comedies such as As You Like It, is undeniably the inclusion of both a dominant and a secondary major female characters. The significance of the secondary female lead is often determined by the skill of the actress portraying that role. The actress can make the character seem somewhat mysterious and forgettable, as seen by the DePaul student’s portrayal of Hero in Much Ado About Nothing, or she can appear as significant and mesmerizing as the lead female. In Griffin’s production of As You Like It this notion of the importance of ctor/ character portrayals is illuminated. For instance, one of the most memorable characters in the entire play is Cecilia, the secondary female character played by Chaon Cross. Cross is able to depict the comedic character almost perfectly through well-rounded acting and clear knowledge of Shakespearean language. In Cecilia’s character, Cross symbolizes a playful, charming, immensely loyal, witty and charismatic supporting female role. This is apparent in her juxtaposition of speech and comedic body language. Crucial plot and thematic elements of As You Like It are depicted by the actors as well. For instance, the review on human life illustrated by the character Jaques, played by Ross Lehman in this production, is witnessed in  Lehman’s acting skills in his performance as Jaques. He emphasizes the faults and ambitions of life in a funny and touching manner by offering both comedic and emotional moments for the audience throughout the play. There are many other characters in the production that portrayed memorable performances illuminating the underlying themes and comedic elements present in the correct use/ knowledge of Shakespearean language and acting. These actors included:   Kate Fry as Rosalind, Matt Schwader as Orlando, Dennis Kelly as Adam, Phillip James Brannon as Touchstone, Matt DeCaro as Duke Frederick, Kevin Gudahl as Duke Senior, and Elizabeth Ledo as Phoebe. Each character possesses unique and intricately detailed costumes and character traits crucial to the understanding of the overall production. The costumes are vivid with detail and evoke the use of imagination and feeling that the audience is witnessing the action of the play within actual setting of certain scenes (whether the orchard, royal court, or forest). The differences in characters’ personality, social class, and hierarchy alongside other characteristic elements are illuminated through the costumes intricate glamour and detail juxtaposed with a more simplistic but still vibrant use of props and set. In addition to individual actor performances, the Chicago Shakespeare Theatre’s production of As You Like It signifies the epitome of Shakespeare’s ’â€Å"glorious romantic comedy† through the use of a unique/ one of a kind setting. The set designs present are an attractive and ubstantial visual supplement to the action taking place on stage at every point throughout the progress of the play. This can be perceived in almost every scene of the production but is most notably present in the opening scene, depicting a lush/ vivid apple orchard with minimalistic use of props, as well as the more luxurious forest setting that accompanies it. The scenery presented by lighting, sound, props hanging from the ceiling, and the perception of depth in the stage prominently adds to the attraction and understanding of the production. The lighting and music of the production present in set/ scene changes enhances the plot developments as well as the overall journey and adventure portrayed in the play’s narrative nature. The set design of various different scenes is thus completely believable and enchanting; employing the audience’s complete inclusion into the imaginative world of the production. Alongside the appearance of setting in the various scenes, the intricately detailed costumes are exquisite and add a visually appealing aspect to the performances of the actors. At the same time, the creativity and uniqueness of each character’s costume mandates the audience’s teleportation from the feeling of watching a traditional play to the feeling that they are in fact present in this magical/ timeless world of As You Like It. Overall, As You Like It, a production shown by the Chicago Shakespeare Theatre, is a professional and glamorous portrayal of traditional/ contemporary Shakespearean comedy. Every aspect of the theatre and production is employed into the narrative element of the play which makes this adaptation of As You Like It unlike any other theatre experience. For instance, the elaborate theatre space is perfect for setting the mood of seeing a professional Shakespearean production and the way in which the stage is the center of vision no matter where you sit in the audience allows sound to travel in a surreal, surround sound manner only enhances this feeling. The actors’ use/ knowledge of Shakespearean language in conjunction with their use of space (entire theatre used as stage while most of the action is mere inches away from the first row of audience members) makes the production flow well while at the same time enhancing the professional feel of the production as well. In this professional nature of the production nearly every aspect and element of Shakespearean comedy is exemplified. Through Gary Griffin’s production of As You Like It, the Chicago Shakespeare Theatre is able to directly exemplify the notion of ideal Shakespearean plays and why these plays still remain so popular and relevant in our society today even four hundred years later.

A Definition Of Collaborative Learning English Language Essay What is collaborative learning? This article looks at the definitions of collaborative learning and provides the reader with an overview of the definitions of collaborative learning, its various forms, and its goals, uses and scope vis-a-vis the traditional classroom and school system. It was midnight on a school night. I saw the light on in my 16 year olds room and went to tell him to go to sleep. His door was uncharacteristically open and I could hear animated voices from inside his room. I found him on Skype with a couple of friends. They were quizzing each other, working out numericals and clarifying each others concepts for a big physics test next morning. Have you worked on your homework math problems with a friend? Did you ever get together with peers in a group to understand a difficult theory in college? Did you ever brainstorm with other classmates on a school project? Weve all done collaborative learning at some stage of our lives; we just didnt know what to call it. The very dictionary definition of collaboration will tell us what collaborative learning means. The word collaboration brings together the Latin col- meaning with or together with the Latin labor or toil. For educationists, collaborative learning is a comprehensive term for a variety of educational approaches involving joint intellectual effort by students, or students and teachers together.1 How can teachers use Collaborative Learning? Collaborative Learning happens when students work together or are made to work together in pairs or groups: to understand or make meaning of a concept or text to create a solution for a problem (given in class or self-discovered) to explore a topic, a question, an area of knowledge to apply the principles learned in their curriculum to conceive of new ways to apply the knowledge they have learned in class to construct a tangible article or a physical object (for example, a report, a term-paper, a model volcano, a recycled-paper bag, a solar panel, an  electric  vehicle) out of the course-learning The above, of course, is an indicative and not a comprehensive list of the ways in which collaborative learning may be used by teachers. Forms of Collaborative Learning Many educators seem to confuse collaborative learning with the more structured approach of cooperative learning. Let us say that cooperative learning is a type of collaborative learning. The developers of cooperative learning models and strategies have laid out several specific components that teachers must control: a small, inter-dependent group determined by the teacher, face to face interaction, carefully structured activity leading to the accomplishment of a predetermined goal, individual accountability of every member of the group and a groups assessment and processing of its own work as a team.2 Other types of collaborative learning that may be used both inside and out of a classroom structure are discussions,  brainstorming  sessions, peer-teaching groups, workshops, team projects, group field-work, study groups, seminars, simulations, role-plays, case-studies etc. Collaborative learning, thus, can be specific, controlled and structured or it can be spontaneous, experiential and totally open-ended. Collaborative Learning in the traditional school system Of course, the traditional school system is at odds with the very spirit of collaborative learning. Schools, as they were conceived and as they are still administered, are essentially authoritarian constructs. Lectures are the preferred method of teaching and the formal physical, emotional and psychological set-up of the traditional classroom is confrontational. Teachers are still equated with discipline and consequences. They are the transmitters of knowledge and the evaluators of the students grasp of the information they have given to the students. They are the regulators of the competition that is encouraged between students through the assessments and recognition and rewards for individual achievement. Conversation is discouraged in traditional classrooms where learning is a solitary pursuit of an individual student faced with the information being given to him from the front of a classroom by a teacher who is the expert instructor. There are definitive syllabi for every subject with course content to be covered in a set period of time; there are lecture plans for every teaching hour and teachers are under pressure to ensure that their students have ingested the carefully planned and delivered information and are capable of regurgitating it as required during formal assessments. Memory is more important than assimilation. In collaborative learning, on the other hand, the process of learning is basically more important than what is learned. Students are taught, by hands-on experience, how to learn and not what to learn. When they learn, assimilation of the material is an absolute requirement as a student must take the material, absorb it, make it his own and then present it or teach it to others. Mere memorizing will not help the collaborative learner. Changing paradigms of an evolving system Collaborative learning, by its very definition, takes the power away from the teacher as guru and distributes that power among the students as self-sustaining, motivated learners who take ownership and responsibility of the entire process of leaning in an interactive, talking-to-each-other and engaged manner. The teacher becomes merely a facilitator, an expert designer of a students intellectual process and a mid-wife of a more emergent learning process.  3 The traditional us versus them power-structure of the traditional school is obviously not conducive to the mutual trust that is required for purely collaborative learning. It is also relevant to point out here that like teachers, students too have to be prepared to take on the challenges and opportunities offered by collaborative learning. I must emphasize that collaborative learning is a tool, just like other teaching methodologies. It is up to the school and the teacher to use this or another tool depending on the objective, the task, the group and the preparedness of the students. A group-discussion may enhance a lecture; it cannot and should not replace it until both teachers and students are ready for the complete shift to another paradigm. As more and more teachers change their classroom strategies and re-orient their relationship to the curriculum from the traditional transmission to transactions that lead to transformations in the personal and social relations of the student to his curriculum,the school system is also slowly evolving and accommodating itself to the more student-centered, process-oriented and non-competitive model that defines collaborative learning. Introduction Science is a creature that continues to evolve at a much higher rate than the beings that gave it birth. The transformation time from plant, to ape, to human far exceeds the time from a calculator to a computer. However science in the past has always remained distant. It has allowed for advances in production, transportation, and even entertainment, but never in history has science be able to so deeply affect our lives as genetic engineering will undoubtedly do. With the new technology there will be, of course, people against it. People who are afraid that genetic engineering and cloning are nothing more than â€Å"toys of the devil†. They fear that it is unsafe. However, I believe genetic engineering is a safe and powerful tool that will yield extraordinary results, specifically in the field of medicine. It will usher in a world where gene defects, bacterial disease, and even aging are a thing of the past. By understanding genetic engineering and it’s history, discover ing it’s possibilities, and answering the moral and safety questions it brings forth, the blanket of fear covering this remarkable technical miracle can be lifted. The first step to understanding genetic engineering and embraccing its possibilities for society is to obtain a rough knowledge base of its history and method. To start off with, you must somehow find an understanding of how individuals pass on characteristics to their offspring. For instance, in regards to eye colour, a child could receive one set of genes from his or her father that were encoded one blue, and the other brown. The same child could also receive two brown genes from his or her mother. The conclusion for this inheritance would be the child has a three in four chance of having brown eyes, and a one in three chance of having blue eyes. Genes are transmitted through chromosomes, which reside in the nucleus of every living organism's cells. Each chromosome is made up of fine strands of deoxyribonucleic acids, or DNA. The information carried on the DNA determines the cells function within the organism. "The new science of genetic engineering aims to take a dramatic short cut in the slow process of evolution". In essence, scientists aim to remove one gene from an organism's DNA, and place it into the DNA of another organism. Reading and Writing Essay

Learning how to read back in 1970 is quite different from learning how to read in the world we live in today. Learning to read was somewhat difficult for me as a child. We didn’t have the fancy reading tools back in the day, such as: Hooked on Phonics or Phonemic Awareness. Looking back over my life, I can remember when I was in the first grade, about six or seven years old and I brought home my first report card. On my report card my teacher pointed out an area of weakness in reading. I can still hear my father saying â€Å"Susie are you having trouble reading? † I would respond as any typical six year old child would do by shrugging my shoulders and saying â€Å"I don’t know. † My father was a concerned parent and wanted to help his child. He didn’t want to see me struggle in school nor did he want to see me fall behind. My father was determined he was going to help me improve my reading skills and by doing so my reading grade would improve also. So therefore, my father set up a strategy plan. Every evening after school my father and I would sit down at the dinner table and work on improving my reading skills. His first step was to see if I could sound out all the letters of the alphabet. Once he realized I was able to sound out all letters in the alphabet, his next step was to determine if I could sound out all the vowels: long, short and house top vowels too. The third step was to begin sounding out small words such as: the, cat, red, run, ran, like etc. Once I began to get comfortable reading on my own while my father prepared dinner for my two brothers and I, he would have me to sit at the dinner table and read aloud to him fifteen minutes each day. Eventually with with all the hard work and dedication my reading skills gradually improved. After all, they say practice makes perfect. As I grew older I started enjoying reading much more than I ever realized. I had to find out what type of books and magazines I enjoyed reading. In other words, I had to find my reading niche. I enjoy reading books like Mama, How Stella got her Grove Back, The Autobiography of Malcolm X, A Day Late and a Dollar Short by Terry McMillan. Reading can be very enjoyable, relaxing and mind stimulating as well. Reading exercises our brain. One can self improve while gaining experience from other people by reading. Reading is knowledge and power. Reading teaches children about the world around them. Reading develops a child’s imagination. Now that I have returned to college, reading and writing is a common part of everyday life. Every semester I have had several different assignments that required reading and writing skills. I have had to do an essay, public speaking presentations which involved reading and writing. Reading is very important because one can explore themselves to new things. One can also self improve while gaining experience from other people. Reading can be used for connecting your brain and it can also boost ones imagination and creativity. Reading exercises ones brain. Many factors play a vital role in a child’s growth and development. Reading develops a child’s imagination. Children who read do better at school. Reading relaxes the body and calms the mind. Reading is a great source of entertainment. This is an important point because these days we seem to have forgotten how to relax and especially how to be silent. In almost everything we do reading and writing is a vital part of our everyday lives.

Fraught Is a Heavy Word â€Å"Fraught† Is a Heavy Word â€Å"Fraught† Is a Heavy Word By Mark Nichol Fraught, one of my favorite words, is fraught with meaning. The term, a variant of freight, was originally a noun and an adjective, with a verb form of fraughten. However, the noun, meaning â€Å"cargo,† disappeared from English (though it survives in Scottish), the verb is entirely extinct, and the adjectival form, which means â€Å"laden, full of† has survived only (with the exception, again, of Scottish) in its centuries-old figurative sense. Now, when a situation is fraught, it is accompanied by, or full of, a feeling usually danger, peril, or some synonymous (and usually sinister) quality. There are exceptions: Note the use in the first paragraph of this post, and a place can be said to be fraught with memories, which may be positive, negative, or a combination thereof. However, because of the word’s often negative connotation, fraught has developed an additional sense of â€Å"uneasy,† and when used this way, it requires no object. For example, one might write, â€Å"We found ourselves in a fraught predicament.† In conversation and in writing, use of the word is fraught with consequences: For one thing, it’s a fairly obscure term, though â€Å"fraught with (blank)† constructions are common enough to form a class of clichà ©s. Furthermore, the use of fraught in the sense of a situation involving emotional turmoil may be unfamiliar to your audience. But fraught is loaded with the strength of precision its meanings are sparse and specific, so it packs a punch and you should hesitate to allow a potent word’s relative rarity to disqualify it from your vocabulary. Properties of Poly(B-amino Ester)s The poly(b-amino ester)s, a class of biodegradable cationic polymers, were firstly  prepared by Chiellini in 198340. These polymers were based on poly(amidoamine)s  developed in 1970 by Ferruti41, that contain tertiary amines in their backbones and  can be synthesized by a simple Michael addition reaction of bifunctional amines  and bisacrylamides. However, the interest over the use of poly(b-amino ester)s rised  significantly after its use as transfection reagent at Langer Lab in 200042. The development  of poly(b-amino ester)s emerged by the need to develop a cationic polymer  for gene delivery with high transfection efficiency and long-term biocompatibility  including hydrolyzable moieties easily degradable into non-toxic small molecule  byproducts. The synthesis of this polymer can easily be accomplished: without  necessity of independent preparation of specialized monomers; the use of stoichiometric  amounts of expensive coupling reagents, or amine protecti on strategies prior  to polymerization42. The main general objective of the work of mentioned research  group was to develop a polymer-based non-viral vector more efficient and less cytotoxic  than other cationic polymers used at that time for this purpose (such as,  polyethylenimine (PEI) or poly(L-lysine) (PLL)). In fact, poly(b-amino ester) approach exhibited a particularly attractive basis for  the development of new polymer-based transfection vectors for several reasons: the  polymers contain the required amines (positive charges to complex genetic material);  readily degradable linkages (by hydrolysis of ester bonds in the polymer backbones  may increase the biodegradability and biocompatibility); and multiple analogues  could be synthesized directly from compounds commercially available (easy and inexpensive  synthesis) allowing to tune polymer properties (like buffering capacity)42. Besides being used as transfection vector, PbAEs has been also applied in others  biomedical areas, such as delivery systems for drugs43;44 or proteins45;46, magnetic  resonance imaging agents47;48, or as scaffold for tissue engineering49;50. Synthesis and main physicochemical properties of poly(b-amino ester)s The poly(b-amino ester)s are easily synthesized by the conjugate addition of a primary  amine or bis(secondary amine) and a diacrylate, in a one-step reaction without  any side product that need be removed through further purification steps. It can be  prepared without solvents, catalysts, or complex protecting group strategies42;51. Depending on the ratio of monomers during the synthesis, poly(b-amino ester)s  can be tailored to have either amine- or diacrylate-terminated chains. An excess of  either diacrylate or amine monomer results in a prevalence of acrylate- or amineterminated  poly(b-amino ester)s, respectively52;53. The synthesis is performed either neat (solvent free) or in anhydrous organic  solvents to mitigate hydrolytic degradation during synthesis42;54. Normally, experiments  using solvents occur at lower temperature and over long periods of time  compared to solvent-free formulations. Table 1.3 summarizes the main reactions for  the synthesis of PbAE and the obtained properties such as molecular weight, polydispersity  index (à ), solvent solubility or yield. The most common solvents used are dimethylsulfoxide (DMSO), chloroform  (CHCl3), or dichloromethane (CH2Cl2)57. However, others solvents have also been  used, such as methanol, N,N-dimethylformamide (DMF) or N,N-dimethylacetamide  (DMA)59;61–63. The solvent used has influence on the final molecular weight of the  PbAE. For example, the use of CH2Cl2 typically yields higher molecular weight  polymer compared to THF42. On the other hand, solvent-free polymerizations maximize monomer concentrations,  thus favoring the intermolecular addition over intramolecular cyclization reaction64. The absence of solvent also allows rising temperature resulting in a higher  reaction rate and a lower viscosity of the reacting mixture, assisting to compensate  the higher viscosity found on the solvent-free systems. The combination between  increased monomer concentration and reaction temperature resulting in a reduction  in the reaction time64. The solvent-free reactions also allows the generation of higher  molecular weight polymers, besides increasing the reaction rate and obviating the  solvent removal step53;64. After polymerization, PbAE can be precipitated, normally in cold diethyl ether,  hexane42, ether65 or ethyl ether58 and/or then dried under vacuum57;65. Frequently, PbAEs are immediately used or stored in the cold conditions (4 _C52;66;67, 0 _C62, or  -20 _C68–70). Some PbAEs should be also kept airproof due to its strong moisture  absorption ability and easy degradation71. Concerning to the biodegradation and biocompatibility, PbAEs have been shown  generally to possess low cytotoxicity and good biocompatibility42;52;61;55;72. Different  studies have suggested that PbAEs are significantly less toxic than currently available  cationic polymers, such as, PEI and PLL51;64. Nevertheless, the increase of the  number of carbons in the backbone or side chain is associated to the increase of the  cytotoxicity73. PbAE degrade under physiological conditions via hydrolysis of their  backbone ester bonds to yield small molecular weight b-amino acids biologically  inert derivatives42;51;55;74. Some results revealed that the degradation rate of poly(b  amino ester)s was highly dependent on the hydrophilicity of the polymer, i.e., the  more hydrophilic the polymer is, the faster the degradation occurs75;76. In Table 1.4 are summarized the main characteristic of PbAEs which make them  a promising polymeric non-viral vector for gene delivery. Combinatorial libraries a fast and efficient way to evaluate different poly(bamino ester)s A fast and efficient way to study the relationships between structure and function  in particular material that could be prepared with different reagents is using combinatorial  libraries. Due to promising preliminar results of PbAEs as non-viral vectors,  Langer research group reported a parallel approach for the synthesis of hundreds of  PbAEs with different structures and the application of these libraries to a rapid and  high throughput identification of new transfection reagents and structure-function trends. For this purpose, major contributions have been reported52;53;57;66;67;72;75;77;78  not only exploring the possible structure/function relationships, but also imposing  an assortment of monomers (amines were denoted by numbers and acrylates by latin  alphabet letters) used in order to facilitate cataloging of different PbAEs (Table 1.5  and Tables A.1 and A.2 (Appendix A)). The first initial library screening was synthesized in 2001 by Lynn51. 140 Different  PbAEs from 7 diacrylates and 20 amines were prepared with molecular weights  between 2,000 and 50,000 g.mol-1. From this, polymers C93 (Mw = 3180 g.mol-1) and  G28 (Mw = 9170 g.mol-1) revealing transfection levels 4-8 times higher than control  experiments employing PEI. At same time, it was observed that for transfection efficiency,  high molecular weight was not an important parameter. This work was then  completed in 2003 by Akinc57, where biophysical properties and the ability of each  polymer/DNA complex to overcome important cellular barriers to gene deliver were investigated. As previous experiments, complexes formed from polymers C93 and  G28, revealed higher levels of internalization compared to †naked† DNA, displaying  18- and 32-fold more internalization, respectively. In contrast, the majority of the  polyplexes were found to be uptake-limited. Regarding d iameter and zeta potential,  out of 10 polymer/DNA complexes with the highest internalization rates, all  had diameters lower than 250 nm and 9 had positive zeta potentials. By measuring  the pH environment of delivered DNA through fluorescence-based flow cytometry  protocol using plasmid DNA covalently labeled with fluorescein (pH sensitive) and  Cy5 (pH insensitive) it was possible to investigate the lysosomal trafficking of the  polyplexes. The results demonstrated that complexes based on polymers C93 and  G28 were found to have near neutral pH measurements, indicating that they were  able to avoid acidic lysosomal trafficking. In the same year, Akinc64 studied the  effect of polymer molecular weight, polymer chain end-group, and polymer/DNA  ratios on in vitro gene delivery. For this purpose, 12 different structures were synthesized  based only in two different PbAE (C28 prepared from 1,4-butanediol diacrylate  and 1-aminobutanol and E28 prepared from 1,6 -hexanediol diacrylate and  1-aminobutanol) (Figure 1.6.) These structures were synthesized by varying amine/diacrylate stoichiometric ratios, resulting in PbAEs with either acrylate or amine end-groups and with molecular  weights ranging from 3,350 to 18,000 g.mol-1. Polymers were then tested, using high  throughput methods, at nine different polymer/DNA ratios between 10/1 (w/w)  and 150/1 (w/w). Concerning terminal groups, it was found that amino-terminated  polymers transfected cells more effectively than acrylate-terminated polymers. In  contrast, none of the acrylate terminated PbAEs mediated appreciable levels of  transfection activity under any of the assessed conditions. These findings suggest that end-chains of PbAE have crucial importance in transfection activity. Concerning  molecular weight effect, highest levels of transfection occurred using the higher  molecular weight samples of both amine-terminated C28 (Mw _13100 g.mol-1 and  E28 (Mw _13400 g.mol-1). Regarding the optimal polymer/DNA ratios for these   polymers, it was observed a markedly difference, 150/1 (w/w) for C28 and 30/1 for  E28. These results highlighted the importance of polymer molecular weight, polymer/DNA ratio, and the chain end-groups in gene transfection activity. Moreover, it  has found the fact that two similar polymer structures, differing only by two carbons  in the repeating unit, have different optimal transfection parameters emphasizing  the usefulness of library screening to perform these optimizations for each unique  polymer structure. Meanwhile, in 2003, Anderson52 described, for the first time,  a high-throughput and semi-automated methodology using fluid-handling systems  for the synthesis and screening of a library of PbAEs to be used as gene carrier. A crucial feature of these methods was that all process of synthesis, storage, and  cell-based assays were performed without removing solvent (DMSO). By using these  methods, it was possible to synthesize a library of 2350 structurally unique, degradable  and cationic polymers in a single day and then test those as transfection reagent  at a rate of _1000 per day. Among PbAEs tested, it was identified 46 polymers  that transfect in COS-7 as good as or better than PEI. The common characteristic  among them was the use of a hydrophobic diacrylate monomer. Moreover, in the  hit structures mono- or dialcohol side groups and linear, bis(secondary amines) are  over represented. From data obtained from this library, Anderson67, in 2004, continued  his study developing a new polymer library of >500 PbAE using monomers  that led higher transfection efficiency in the previous studies and optimizing their  polymerization conditions. The top performing polyplexes were asses sed by using  an in vitro high-throughput transfection efficiency and cytotoxicity assays at different N/P ratios. As previously observed, the most promising polymers are based on  hydrophobic acrylates and amines with alcohol groups. Among those, C32 stood  out due to higher transfection activity with no associated cytotoxicity. The efficiency  to deliver DNA was evaluated in mice after intra-tumoral (i.t.) and intra-muscular  (i.m.) injection. The results revealed important differences. While by i.t injection  C32 delivered DNA 4-fold better than jetPEI R , a commercial polymeric non-viral  vector, by i.m. administration transfection was rarely observed. C32 was then assessed  for DNA construct encoding the DT-A (DT-A DNA) deliver to cells in culture  and to xenografts derived from androgen-sensitive human prostate adenocarcinoma  cells (LNCaP). Results showed that DT-A DNA was successfully delivered and the  protein expressed in tumor cells in culture. In hu man xenografts, the growth was  suppressed in 40% of treated tumors. The fact of C32 is non-toxic and it is able to  transfect efficiently tumors locally and transfects healthy muscle poorly turned it as  a promising carrier for the local treatment of cancer. From here, a panoply of results based in PbAE combinatorial library appeared. In  2005, Anderson53, prepared a new library of 486 second-generation PbAE based on  polymers with 70 different primary structures and with different molecular weights. These 70 polymers were synthesized using monomers previously identified as common  to effective gene delivery polymers. This library was then characterized by  molecular weight of polymers, particle size, surface charge, optimal polymer/DNA  ratio and transfection efficiency in COS-7 of polymer/DNA complexes. Results  showed that from 70 polymers with primary structures, 20 possess transfection activities  as good as or better than Lipofectamine R 2000, one of the most effective commercially  available lipid reagents. Results also revealed that, in general, the most  effective polymers/DNA complexes had In 2006, Green79, synthesized, on a larger scale and at a range of molecular  weights, the top 486 of 2350 PbAEs previously assessed52 and studied their ability to  deliver DNA. These PbAEs were tested, firstly, on the basis of transfection efficacy in  COS-7 cells in serum-free conditions, and then, the 11 of the best-performing PbAEs  structures were further analyzed. The transfection conditions were optimized in human  umbilical vein endothelial cells (HUVECs) in the presence of serum. In this  study, the influence of the factors like polymer structure and molecular weight, and  biophysical properties of the polyplexes (such as, particle size, zeta potential, and  particle stability throughout time) were studied. The results showed that many of  the polyplexes formed have identical biophysical properties in the presence of buffer,  but, when in the presence of serum proteins their biophysical properties changed differentially,  influencing the transfection ac tivity. Concerning to the size, the results  showed that in spite of all vectors condensed DNA into small particles below 150 nm  in buffer, only a few, such as C32, JJ32 and E28, formed small (_200 nm) and stable  particles in serum. C32, JJ32 and E28 revealed also high transfection activity both  in the absence of serum in COS-7 cell line as in the presence of serum in HUVEC  cell line. Moreover, C32 transfected HUVECs in the presence of serum significantly  higher than jetPEI R and Lipofectamine R 2000, the two top commercially available  transfection reagents. The 3 mentioned PbAEs share a nearly identical structure. The acrylate monomers of these polymers, C, JJ, and E, differ by only their carbon  chain lengths (4, 5, and 6 carbons, respectively). Similarly, amines 20, 28, and 32  differ also by only the length of their carbon chain (3, 4, and 5 carbons, respectively). For example, polymers prepared with the same acrylate monomer (C) in which itwas increased the length of the carbons chain of the amine monomer resulted in  an increased transfection efficacy (C32 (5 carbons) > C28 (4 carbons) > C20 (3 carbons))  of these polymers-based polyplexes. Interestingly, this study reinforced C32  as the lead PbAE vector and revealed other potential two, JJ28 and E28, which previously  showedto be poor vectors. On the other hand, C28 and U28, previously  recognized as an efficient transfection reagent, were found to transfect inefficiently  HUVEC in serum. By constructing a new library of end-modified PbAE, the research  was continued78 in order to understand the structure-function relationship  of terminal modification of PbAE in transfection activity. For this purpose, it was  used twelve different amine capping reagents to end-modify C32, D60 and C20. The  choice of these 3 PbAEs was based in their transfection activity: C32, the most effective; D60, an effective transfection reagent with a significantly different structure  from that of C32; and, C20, a poor transfection reagent but with similar structure  to C32 differing only in the length of the amine monomer. The results showed  that some PbAEs-based vectors (C32-103 and C32-117) were able to deliver DNA by  approximately two orders of magnitude higher than unmodified C32, PEI (25,000  g.mol-1) or Lipofectamine R2000, and, at levels comparable to adenovirus at a reasonably  high level of infectivity (multiplicity of infection = 100). Once again, it was  demonstrated that small structural changes influence greatly gene delivery, from biophysical  properties (such as, DNA binding affinity, particle size, intracellular DNA  uptake) until final protein expression. From these 3 polymers assessed, C20 was the  one who transfected cells much less effectively, although it has seen a remarkably  improvement with end-modifications. As expected, C 32-based polyplexes, based on  C32-103 and C32-117, revealed the higher transfection efficiency enhancing cellular  DNA uptake up to five-fold compared to unmodified C32. Interestingly, and in a  general way, terminal modifications of C32 with primary alkyl diamines were more  effective than those with PEG spacers, revealing that a degree of hydrophobicity at  the chain ends is an added value for these polymers. Another interesting fact in terminal  modification of C32 was that at least a three carbon spacer between terminal  amines is necessary to obtain an efficient gene delivery. For example, results showed  that C32-103 transfection efficiency is 130- and 300-fold higher than C32-102 on the  COS-7 and HepG2 cell lines, respectively. As the molecular weight was the same,  this result demonstrated the critical role of the chain ends in transfection activity. In order to better understand the role of the chain ends in transfection efficiency  a new library of end-modified C32 was synthesized by Zugates80 in 2007 using 37  different amine molecules to end-modify the PbAE. In a general way, it was observed  that polymers end-capped with hydrophilic amine end groups containing  hydroxyls or additional amines led to higher transfection efficiency. On the other  hand, terminal-modifications with hydrophobic amines containing alkyl chains or  aromatic rings proved to be much less effective. Bob Dylan's, ';The Times They Are A-Changin'; is an anthem for the oppressed, down-trodden young people, while warning that oppressors and abusers will be victims of their own actions. In the beginning of the poem, Dylan speaks to everyone and talks of the change coming from young people who feel that laws from the government and mom and dad's rules are smothering. He emphasizes 'everyone'; by using water to help the reader visualize how complete the wave of change will surround people. He then uses the water in a sink or swim analogy illustrating the direness of the situation. Throughout the poem, he shows the extent and gives examples of who will be affected. Although he draws class lines and social standings, the opportunity to change along with the times is always present. Dylan points specifically to 'senators, congressmen,'; ';mothers and fathers,'; because they have the most influence on America's youth. Dylan calls on the American government to 'Please heed the call' which shows that in the beginning, respect and persuasion will be used. The next two lines begin 'Don't'; which indicates a stronger will and mind set. 'For he that gets hurt/Will be he who is stalled,'; illustrates that if there is resistance to young people's ideas against the war in Vietnam, the idea of free love and the distaste for accepted social structures, that peace may not be an option. Sir Gawain and the Green Knight :: Sir Gawain Green Knight Poem Essays

Sir Gawain and the Green Knight Sir Gawain and the Green Knight was written by an anonymous author in the 14th century. It was written in a dialect from Northern England. The poem uses alliteration similar to the Anglo-Saxon form of poetry. Alliteration uses a repetition of consonants. The poem ends the way it begins. At the end of each scene, the section of the poem concludes with a sharp rhyme. There are many patterns that are developed by the author in the poem. There are three literal hunts with the deer, boar and fox. The deer is represented as being timid; the boar is represented as being ferocious, and the fox as being cunning and sly. There are also three symbolic hunts, three attempts to seduce Gawain and the three hits of the Green Knight's ax. The poem seems to emphasize magical elements such as Morgan, Merlin, and the Green Knight. The poem gives us a very realistic depiction of the slaughtering of the animals, the big feasts that take place, as well as the changing seasons. The poem also contains many religious elements such as the celebrations that take place according to the church calendar. King Authur's court is in the midst of a celebration that has taken place for fifteen days. They are celebrating the New Year, and in the middle of their celebration, the Green Knight and his green horse come barging into the hall. Sir Gawain is the youngest knight of the Round Table. He is the only one that volunteers to play the "game" with the Green Knight. The game is that the challenger gets a chance to hit the Green Knight now, but in a year and a day, he must go to the Green Knight's chapel and then the Green Knight will test him and if he fails the tests, the Green Knight will hit him. The Green Knight has no fear, and he even lowers his neck to make it easier for Gawain. Gawain bravely cuts off the Green Knight's head, and the Green Knight proceeds to pick it up. Then the head speaks to the people on the dais, and he reminds them of the deal. The Green Knight then rides off on his horse with his head in his hand. The people in the hall are surprised that King Arthur allows Gawain to go on this journey to the Green Knight's chapel. Evaluation of the iPhone 5S

In the first 24 hours of the launch on September 20, 2013, Apple Inc. sold 2,750,000 iPhone 5s'. The Apple iPhone is the top sold smartphone and is continuing to control the market. The iPhone 5s is a very respected and prominent product, it is used by millions of people for many tasks like work and school. There are few things that stand out to me that I have chosen for the iPhone 5s to evaluate it in: Design, Technology, Software, and the App Store. I believe that the iPhone 5s proves itself in all of these categories, and many people would agree with me, as well as disagree.The design plays a huge role in cell phones, it has to grasp the buyers attention and make them want the product just by looking at it or by watching someone use it. The iPhone 5s does this really well, Apple engineers and designers managed to compress the technologies they made inside a space that’s a mere 7.6 millimeters thin and 112 grams light. This redesign resulted in an incredibly thin, impressive ly light, extraordinarily powerful smartphone. The engineering challenge was significant, but they succeeded in adding more to it without making iPhone 5s bigger or heavier.The iPhone began with something perfectly simple, your finger. And now Apple took touch to the next possible place with Touch ID, the fingerprint identity sensor. Your fingerprint is the perfect password, you always have it with you and no one can ever guess what it is. But beyond that, it just made sense that your phone should recognize you. It should learn you, not require you to memorize and enter passwords in order to use it. Apple also put the sensor in the best place possible, where you most commonly rest your thumb, on the Home button. It is amazing how Apple increased the speed of the iPhone 5s without decreasing the battery life, in fact, they increased the battery life.This is all true because Apple engineered a new processing chip. The A7 chip is designed around 64-bit architecture, which is a first fo r any smartphone. The move to 64-bit chip made it possible for desktop-class processing power in the palm of people’s hands. The A7 chip gives iPhone 5s the power it needs for all the capabilities it now has. It’s up to 2x faster than the previous generation in both CPU and graphics performance, it has an all-new image signal processor, and it supports OpenGL ES 3.0, which enables visual effects previously possible only on computers and gaming consoles.Even with all these performance enhancements,  A7 is still energy efficient.The new M7 coprocessor is designed to make iPhone 5s even more efficient. It offloads work from the A7 chip by collecting motion data, then Apps can use this data without constantly using the A7 chip. Because M7 is engineered for this specific task, it uses significantly less power than A7 would require, so battery life is spared.Another big role in cell phones is its features, the more there are, the more appealing it is. Apple is an outstandi ng contender in this specific criterion because they make their own advanced technology, which adds many cool feature. The iPhone 5s is precision crafted down to the micron. It has a beautiful aluminum housing, very sleek metal and glass, sapphire crystal in the Home button and more sapphire crystal protecting the iSight camera. Design and construction of this level is unmatched so the iPhone 5s looks and feels unbelievably thin and light. It is also available in three elegant colors: gold, silver, and space gray.The most prominent feature of them all is the Touch ID allowing you to use your fingerprint for your password. The iPhone 5s features advanced technologies custom designed for the iSight camera’s hardware and software. Some of these features include: larger pixels, larger sensor, continuous burst mode, true tone flash, auto image stabilization, slow motion video, live video zoom, square photos, panorama photos, and photo filters. Another cool feature is FaceTime, whi ch allows you to video chat with anyone with an Apple product anywhere in the world.The iPhone 5s' LTE is ultrafast allowing it to download and upload data at unimaginable speeds, the iPhone 5s has the fastest LTE speed for any smartphone. The App Store is a fabulous feature, it is easy and fun to use. When downloading an app, all you have to do is scan your finger on the home button and voila, it starts downloading. iCloud is a great feature because it allows you to have everything you need, anywhere you need it. iCloud allows you to have the same data between different Apple devices, which comes in handy for school if you forget something on your iPad so you can bring it up on your iPhone.Software is very important for any smartphone, and with the iPhone 5s, comes iOS 7. iOS 7 is made by Apple Inc.'s workers. They made it simpler, more useful, and more enjoyable while making it instantly familiar. Apple made  iOS 7 very simple and beautiful, they offer the right things, in the r ight place, at the right time. It seems as if it â€Å"just works† all the time, when you pick it up you already know how to use it, that's simplicity. It makes sense why they put certain features in, they put it in when it's truly useful, like the notification center or the control center.It becomes more to you than just a device, iOS 7 invites that kind of connection. Interactions are dynamic, and animations are cinematic. The experience is lively and spirited in so many different ways you wouldn't have expected. Open the Weather app, for example, and you’ll instantly understand. Hail bounces off text, and fog passes in front of it. Storm clouds come into view with a flash of lightning. Suddenly, checking the weather is like looking out a window. iOs 7 also adds a sense of dimension with several layers on the screen tilting according to how you hold it.Apps are a very important part of Apple. It makes for a great experience and fun to use with it's built in apps made by Apple. The built in apps consist of Camera, Photos, Music, Safari, Maps, Siri, Phone, FaceTime, etc. There are a total of 39 built in apps that are great from music to school. The App Store has the best selection of mobile apps, from Apple and third-party developers and they’re all designed specifically for iPhone. The more apps you download, the more you’ll realize your iPhone can do just about anything you can imagine. You can easily find apps that are relevant to your current location and browse categories of apps based on your child’s age and let the App Store update all your apps for you.Purchasing apps is now as easy as placing your finger on your iPhone. Say there’s an app you’re ready to download, with Touch ID, you simply touch the fingerprint identity sensor on the Home button, and in an instant, the App Store recognizes you and approves your transaction. So you don't need to type your Apple ID or remember your password anymore, which is amazingly easy. Shopping in the App Store is a great experience too because it’s easy to find the apps you want, and to discover new apps you didn’t even know you wanted. Browse freely by category, or shop collections of apps and games handpicked by experts. Apple reviews everything on the App Store to guard against malware, so you’re buying and downloading from a trusted source.Overall, the iPhone 5s has a great design, it has many amazing, new features, it's new software is simple and â€Å"just works† and it has plenty of apps made just for the iPhone and its technological abilities. The iPhone 5s, in my opinion, not only meets, but exceeds the criteria for a good smartphone. It is the fastest, most luxurious, easiest to use, and most technologically advanced phone on the market today.
